A-DNA

A-DNA ist eine der möglichen Doppelhelix-Strukturen von DNA.
Eigenschaften
A-DNA ist eine rechtsgängige doppelsträngige DNA-Doppelhelix, die im Vergleich zur B-Form kompakter ist und deren Nukleinbasen nicht orthogonal zur Helixachse ausgerichtet sind. A-DNA ist die bei niedriger Feuchtigkeit vorliegende Form doppelsträngiger DNA, z. B. bei getrockneter DNA oder DNA-Kristallen.[1] Andere DNA-Strukturen sind z. B. B-DNA, Z-DNA, C-DNA, hairpin, triplex, cruciform, left-handed Z-form, tetraplex und A-motif.[2] Möglicherweise wird die A-Form auch bei hybriden DNA-RNA-Doppelhelices ausgebildet, da eine ähnliche Struktur auch die häufigste Form von RNA-RNA-Doppelhelices ist. Die Zuckereinheiten haben bei A-DNA eine 3’-endo-Konformation, im Gegensatz zur B-DNA, welche eine 2’-endo-Konformation der Zuckereinheiten besitzt. Dies ermöglicht es auch doppelsträngigen RNA-Regionen eine A-DNA-ähnliche-Form einzunehmen, da die sterische Hinderung durch die 3’-endo-Konformation nicht bei der Ausbildung der A-Form stört, im Gegensatz zur 2’-endo-Konformation der B-Form. Aufgrund der kompakten Form besitzt A-DNA im Vergleich zu B-DNA eine höhere Anzahl an Nukleinbasen pro Windung der Helix, eine tiefere große Furche und eine flachere kleine Furche. A-DNA ist im Vergleich zu B-DNA um etwa 30 % verkürzt und breiter.
