Cy7 ist eine chemische Verbindung aus der Gruppe der Methinfarbstoffe. Zwei substituierte Indolin-Reste sind über eine Heptamethingruppe verbunden.

Gewinnung und Darstellung

Cy7 kann durch eine mehrstufige Reaktion ausgehend von Phenylhydrazin-4-sulfonsäure[S 1] und 3-Methylbutanon synthetisiert werden.[2]

Eigenschaften und Verwendung

Cy7 ist ein kristalliner Feststoff, der löslich in organischen Lösungsmitteln ist. Die Löslichkeit von Cy7 in Ethanol beträgt etwa 5 mg/ml und in DMSO und Dimethylformamid etwa 10 mg/ml. Es ist ein Cyanin-haltiges Fluorochrom und weist eine maximale Anregung/Emission bei einer Wellenlänge von 745/800 nm auf. Die Verbindung und seine Derivate werden häufig an sekundäre Antikörper konjugiert, um es in der Fluoreszenzmikroskopie und Durchflusszytometrie einzusetzen.[3][4][5][6] Die Indotricarbocyanin-Struktur von Cy7 ist chemisch nicht so stabil wie die Indodicarbocyanin- und Carbocyanin-Farbstofffluorophore von Cy5 und Cy3. Dies erschwert die Herstellung von Cy7-Markern und führt zu einer relativ kurzen Haltbarkeit.[7]
