TRAIL(TNF-related apoptosis-inducing ligand)もしくはTNFSF10(TNF superfamily member 10)、CD253(cluster of differentiation 253) は、アポトーシスと呼ばれる細胞死過程を誘導するリガンドとして機能するタンパク質である[5][6][7]。
TRAILは、大部分の正常組織において産生・分泌されている一方で、腫瘍細胞に対して選択的にアポトーシスを引き起こすサイトカインである[7]。TRAILとその受容体は1990年代半ばから抗がん剤開発の標的となっている(マパツムマブ など)一方で、2013年時点で有意な延命効果が示されたものはまだない[8]。TRAILはさまざまな肺疾患において発症因子もしくは保護因子として示唆されており、特に肺動脈性肺高血圧症の病理において重要である[9]。
遺伝子
ヒトにおいてTRAILをコードしているTNFSF10遺伝子は3番染色体3q26領域に位置し、他のTNFファミリーのメンバーの遺伝子とは近接していない[5]。TNFSF10遺伝子の長さは約20 kbで、5個のエクソンと4個のイントロンから構成される。FasLやTNF-αといった他のTNFファミリーの遺伝子とは対照的に、プロモーター領域はTATAボックスやCAATボックスを欠いており、GATA、AP-1、C/EBP、Sp1といった転写因子の結合が推定されるエレメントが存在している[10][11]。TIC10やONC201といった低分子によってTNFSF10遺伝子の発現をアップレギュレーションすることで、一部のがん細胞の死が誘導されることが報告されている[8][12]。
構造
TRAILは281アミノ酸から構成され、II型膜貫通タンパク質の特徴を有する。N末端側の細胞質ドメインは他のTNFファミリーとの保存性はみられないが、C末端側の細胞外ドメインは保存されており、タンパク質分解によって細胞表面から切断遊離する。TRAILはホモ三量体を形成し、3個の受容体分子に結合する[10]。
機能
TRAILはデスレセプターDR4(TRAIL-RI)およびDR5(TRAIL-RII)に結合し、アポトーシスを誘導する[13][14]。アポトーシス過程はカスパーゼ-8依存的である。カスパーゼ-8は、プロカスパーゼ-3、-6、-7などの下流のエフェクターカスパーゼを活性化し、特定のキナーゼの活性化をもたらす[15]。TRAILはDcR1やDcR2にも結合する。DcR1は細胞質ドメインを持たず、DcR2はデスドメインが切り詰められている。DcR1はTRAILを中和するデコイ受容体として機能する。DcR2の細胞質ドメインは機能的であり、NF-κBを活性化する。DcR2を発現している細胞では、TRAILのDcR2への結合によってNF-κBが活性化され、細胞死シグナル伝達経路に拮抗したり、炎症を促進したりすることが知られている遺伝子の転写が引き起こされる。これらデスレセプター(DR4、DR5)やデコイ受容体(DcR1、DcR2)に対してさまざまな親和性を有するよう改変したリガンドを投与することで、細胞死の1型/2型経路の活性化や一細胞レベルでの変動を制御した、がん細胞の選択的標的化の実現の可能性がある。こうしたアプローチのために、TRAILを模倣した発光性イリジウム錯体ペプチドハイブリッドが合成されている。この人工的TRAIL模倣体はがん細胞上のDR4/DR5に結合し、アポトーシスとネクローシスの双方を介して細胞死を誘導することが示されており、抗がん剤開発の候補分子となっている[16][17]。
創薬標的としてのTRAILとその受容体
TRAILを結合するデスレセプターを標的としたさまざまな薬剤の開発が行われているが、臨床試験においてこうした薬剤に応答するのはがん患者はわずかである。多くのがん細胞ではTRAILに対する抵抗性が獲得されており、TRAILを標的とした治療の効果は限定的なものとなっている[18]。
