TRPM4(Transient receptor potential cation channel subfamily M member 4、TRPチャネルサブタイプM4)はメラスタチン-4(melastatin-4)とも呼ばれる蛋白質であり、遺伝子TRPM4 でコードされる[1][2]。心筋細胞の表面に発現しており、心筋の電気活動に関与している。
このチャネルを薬物で阻害すると心筋梗塞の進行を抑制することが知られている[3]。
TRPM4はNMDA型グルタミン酸受容体(NMDAR)を構成するGluN2A, GluN2Bと結合する[4]。この相互作用はNMDARおよびTRPM4のチャネル機能には影響しないが、この相互作用がNMDA受容体を介した細胞死(興奮毒性)を引き起こす。相互作用はTRPM4のTwinFドメインおよびGluN2のI4ドメイン間で起きており、TRPM4側に結合し相互作用を阻害する薬剤が開発されている。
