Rhodamine is een familie van organische fluorescerende kleurstoffen. De meeste rhodamines zijn kationische verbindingen, bestaande uit kern van xantheen met daaraan amine- en iminegroepen, waardoor ze goed oplosbaar zijn water en alcoholen. Qua structuur zijn ze verwant aan andere kleurstoffen zoals fluoresceïne en eosine. Rhodamines werden door hun opvallende kleur al in de negentiende eeuw beschreven.
De ringstructuren geven de verbindingen sterke fluorescentie. De meeste rhodamines absorberen in het groene tot gele deel van het spectrum en zenden licht uit in het rode deel. Door hun hoge kwantumopbrengst en relatief goede fotostabiliteit zijn rhodamines bijzonder geschikt voor gebruik als fluorescente markers. Voorbeelden van veelgebruikte rhodamines zijn sulforhodamine B, rhodamine WT, rhodamine B, rhodamine 6G en rhodamine 123.
Toepassingen

Rhodamines worden breed toegepast in wetenschappelijk onderzoek en technologie. Afhankelijk van de aanwezige functionele groepen rondom de xantheenkern en het tegenion (chloride of bromide) vertonen rhodamines verschillende fotofysische eigenschappen, zoals absorptie- en emissiemaxima.[1] De stoffen spelen een belangrijke rol in kleurstoflasers (dye lasers).
In de biologie en geneeskunde worden derivaten ervan gebruikt als fluorescente labels voor eiwitten en celstructuren, bijvoorbeeld in fluorescentiemicroscopie en flowcytometrie. In de fysische chemie en milieuwetenschappen worden rhodamines toegepast als tracers voor onderzoek naar stroming en diffusie.[2]
Veiligheid
Sommige rhodamines vertonen toxiciteit en zijn persistent in het milieu. Het gebruik ervan in consumentenproducten is hierdoor beperkt of geheel verboden. In het verleden werd rhodamine gebruikt als kleurstof voor textiel en inkten. Rhodamine WT is een variant speciaal voor milieutesten vanwege lagere toxiciteit.
